Kerman province is the most major pistachio‐growing region in Iran. During 2015–2016, pistachio cultivars were surveyed in the province to detect HSVd. A total of 106 asymptomatic and symptomatic leaf samples showing mosaic, yellowing, and chlorosis symptoms were collected from pistachio trees and assayed for the HSVd infection by reverse transcription polymerase chain reaction, and dot‐blot hybridization. Results showed that four pistachio cultivars (Momtaze, Kalleh‐Ghouchi, Ohadi, Ahmad-aghaei) are infected by HSVd, while no HSVd infection was detected in the Akbari cultivar. In this study, bioassay tests including the mechanical transmission of viroid to pistachio rootstocks were used to evaluate the biological characteristics of Iranian HSVd pistachio variants. The infectivity of one HSVd variant (Ker.Ana.P1) was further verified by mechanical inoculation. The viroid variant was transmitted to two different healthy pistachio rootstocks (Badami, University of California Berkeley I (UCB-1)) using infected sap by three inoculation methods i.e. slashing, rubbing, and RNA injection. Results indicated that all three methods cause infection of the pistachio rootstocks with HSVd. However, the slashing inoculation showed higher transmission efficiency in Badami and UCB-1 at the rate of 86.67 and 80 percent, respectively. According to this study, it seems that HSVd infection is increasing on pistachio trees in Kerman province due to lack of attention to sanitary measures during agricultural operations such as pruning, which leads to transmission of disease agents like viroids.}, keywords = {Biological indexing,Pistachio cultivars,Prokaryotes}, title_fa = {پراکنش ویروئید کوتولگی رازک در باغ‌های پسته استان کرمان و ارزیابی انتقال مکانیکی و برهمکنش دو رقم پایه پسته نسبت به این ویروئید}, abstract_fa = {ویروئید کوتولگی رازک (Hop stunt viroid, HSVd) تعداد زیادی از درختان میوه مانند انگور، مرکبات، آلو، هلو، انجیر، توت، گلابی، پسته و بادام را آلوده می‌نماید. طی اردیبهشت 1393 تا شهریور 1395 تعدادی از باغ‌های پسته استان کرمان به‌عنوان مهمترین منطقه پسته‌کاری کشور، به ‌منظور آلودگی به ویروئید HSVd بررسی شدند. در مجموع 106 نمونه جهت تعیین آلودگی به این ویروئید از برگ‌های درختان پسته با علائم موزائیک، زردی، رنگ‌پریدگی و تعدادی نیز بدون علائم جمع‌آوری شده و توسط آزمون‌های RT-PCR و هیبریداسیون نقطه‌ای ارزیابی گردیدند. نتایج به‌دست آمده آلودگی چهار رقم پسته کله‌قوچی، اوحدی، ممتاز و احمد‌آقایی را به HSVd تأیید کرد، اما در نمونه‌های رقم اکبری آلودگی مشاهده نگردید. جهت آزمون‌های زیست‌سنجی از واریانت ایرانی Ker.Ana.P1، جدا شده از درختان پسته منطقه انار، که قادر به ایجاد آلودگی و بیماری‌زایی روی گیاهان محک (ارقام خیار شناساگر) بود، استفاده گردید. بر‌ اساس نتایج آزمون‌های زیست‌سنجی شامل انتقال مکانیکی واریانت HSVd، از طریق روش‌های سایشی، برشی و تزریق RNA ویروئید به پایه‌های بادامی و University of California Berkeley-I (UCB-1) مشخص گردید که هر سه روش سبب آلودگی پایه‌های پسته می‌گردند و از بین آن‌ها، روش مایه‌زنی برشی، بالاترین راندمان انتقالHSVd  با 67/86 و 80 درصد در پایه‌های بادامی و UCB-1 را نشان داد. بر‌ اساس نتایج بدست آمده در این تحقیق، به‌نظر می‌رسد که آلودگی درختان پسته در استان کرمان نسبت به HSVd به‌دلیل عدم توجه به مسائل بهداشت زراعی و انتقال عوامل بیماری‌زا نظیر ویروئیدها در حین عملیات باغی مانند هرس رو به افزایش می‌باشد.}, keywords_fa = {ارقام پسته,آزمون‌های زیست‌سنجی,پروکاریوت‌ها}, url = {https://pistachio.vru.ac.ir/article_131581.html}, eprint = {https://pistachio.vru.ac.ir/article_131581_7cb32c2cabedb0fd5ddbf70b578678d0.pdf} }
